Liberty Trike on order , I would like to improve the ride, by replacing the rear tires/wheels that are now 16/2.5 with 20/4.0 any thoughts?

I'm not sure that will have the desired effect. You'd likely have to change out all three wheels and tires to make it work. Fat tires have a larger outside diameter to begin with, add larger wheels and the frame of the trike will be jacked up 4 plus inches in the back. Changing geometry of any bike is very tricky to get right and safe. You could make the ride more uncomfortable and dangerous. If you determine the ride isn't what you'd hope, try adjusting air pressure down to minimum specs. Change out the saddle/seat. Add seat-post suspension. Or get slightly larger balloon tires for the stock wheels.
 
My wife has a Liberty Trike. I have considered this same approach to improve the ride, however due to the narrow width of a Liberty we have backed off. Any trike will be a bit tippy until you get use to it. We do adjust the air pressure depending on the riding surface. We also installed a suspension seatpost and changed the seat.

I would ride the trike for a few miles first. My wife sometimes feels uncomfortable if a road has a steep crown.

My thinking is by adding a 4.0 rear tires, it would add a little suspension and a little less tippy
Also add a little balance on the front wheel it might keep the front wheel
16/2.5. 750 w motor from over spinning

I like what J.R. wrote, especially about adding a suspension seat post. It's simple and safe. I have a Kinet Body Float seat post on both of my ebikes (non trike ) and it's
great. I love it ! The Body Float is a little pricey compared to some the other brands, but for me it's worth it. To save some $$ the non carbon fiber Body Float model is a good choice.
