"Best" depends on what you are using your ebike for. I use mine for errands and exercise (which I like to combine). I have a throttle, but only use it to test whether my rear wheel is rubbing on the fender by tipping the bike on the kick stand and throttling. Part of the reason is the throttle is available only on zero pedal assist, and I always want to use pedal assist. Some people use throttle to make quick starts from a stop, but mine won't work that way unless I turn off PAS.
One down side of using throttle exclusively is battery depletion. When you add your own leg wattage, battery life goes way up. I'm sure others will weigh in with a variety of opinions.
My wife and I are pedal-assist only. We like having full control and the “guaranteed” exercise of peddling. We had 2 incidents during group rides where throttle bikes got away from the riders during maneuvers around trail hazards. Luckily only minor injuries to both riders.
My bike has both throttle and pedal assist on all settings except "no assist", in which case it is throttle-only. There's no difference between the two in terms of power or speed—it's the same motor and the same battery. I would never buy a bike without a throttle, and I don't need people who aren't me to decide I don't need a throttle.
