While variable regen is the ultimate tool (usually on a lever), a good controller gives ramping curve adjustments so that you can adjust how it will ramp up when you trigger regen and ramp down when you release it.

Having a twist throttle on the right on my e-bike, I have also put the Front brake on the right.
This is the logic I was use to on motorcycles all my life, and it also make sense as you don't want to be twisting that throttle and braking at the same time.
The ideal configuration for High power is a dual drive train like on the LMX or Boxxbike.
That does require some customization and may not be feasible on your setup though.
You should ask the manufacturer what the spoke tension should be.
If the rim is centered on the HUB, the tension would be the same on both side, but if the rim is off center (asymmetric lacing), then the tension would be different.
